چکیده

The MAGE genes encoding tumor rejection antigens recognized by < 11,s are expressed at the mRNA level in various cancers and in the testis but not in the other normal tissues. The expression of MAGE-1 or MAGE-4 protein in the testicular cells was studied immunohistochemically with the antibodies to the recombinant MAGE-1 or MAGE-4 pro tein. Both proteins were identified in the nucleus and cytoplasm of spermatogonia and in primary spermatocytes but not in spermatids or Sertoli's cells. Therefore, MAGE proteins are normal tissue antigens compartmentalized in particular testicular cells playing an important role in the early phase of the spermatogenesis.

Expression of MAGE-A1, MAGE-A3/4 and NY-ESO-1 cancer-testis antigens in fetal testis.

Immunohistochemical expression of MAGE-A1, MAGE-A3/4 and NY-ESO-1/LAGE-1 cancer testis antigens (CTA) was assessed in 24 fetal testes from 15th to 36th week of gestation. Three monoclonal antibodies were used for immunohistochemical staining: 77B recognizing MAGE-A1, 57B recognizing multiple MAGE-A CTA, and D8.38 recognizing NY-ESO-1/LAGE-1. Expression of MAGE-A1 was not observed in fetal testi...

The mouse Mageb18 gene encodes a ubiquitously expressed type I MAGE protein and regulates cell proliferation and apoptosis in melanoma B16-F0 cells.

Although many cancer vaccines have been developed against type I MAGE (melanoma antigen) genes owing to their shared tumour-specific expression properties, studies about their expression and functions are relatively limited. In the present study, we first identify a non-testis-specific type I MAGE gene, Mageb18 (melanoma antigen family B 18). Mouse Mageb18 is also expressed in digestion- and im...

Identification of a new MAGE gene with tumor-specific expression by representational difference analysis.

Human genes expressed exclusively in tumors and male germ line cells, such as those of the MAGE, BAGE, and GAGE families, encode antigens recognized by T lymphocytes, which are potentially useful for antitumor immunotherapy. To identify new genes of this type, we generated cDNA populations enriched in sequences expressed only in testis and melanoma, using the representational difference analysi...

Cancer Immunity 2:11 (2002) - ARTICLE

Cancer-testis (CT) genes are expressed in a variety of human cancers, but not in normal tissues except for testis, and represent promising targets for immunotherapy and gene therapy. We investigated the expression of 10 CT genes (MAGE-1, MAGE-3, MAGE-4, GAGE, NY-ESO-1, SSX-1, HOM-MEL-40/SSX-2, SSX-4, HOM-TES14/SCP-1, and HOM-TES-85) in 21 hepatocellular carcinoma (HCC) biopsy specimens. The mos...
